The Facts

  • Wake Forest University researchers studying axolotls, zebrafish and mice have identified SP6 and SP8 transcription factors as conserved genetic drivers of appendage regeneration across species. Their research findings were published in the Proceedings of the National Academy of Sciences on April 14.
  • Using CRISPR technology, they removed SP8 from axolotl genomes, halting limb bone regeneration. Deleting SP6 and SP8 from the mouse basal epidermis produced the same result, triggering an IL-17-mediated osteoclast response that degraded remaining tissue instead of rebuilding it.
  • They then engineered an adeno-associated virus carrying a zebrafish tissue regeneration enhancer to deliver FGF8 to amputation sites in mice. The therapy partially restored digit regeneration in SP gene knockout mice and accelerated regrowth in wild-type mice.

SP genes — active in axolotls, zebrafish and mice — form a universal blueprint that humans still carry but don't use.
